Learn what Tyloxapol is. Discover the mechanism, advantages, and applications of the non-ionic surfactant Tyloxapol in eye drops, aerosols, and nano systems.
Tyloxapol is a non-ionic surfactant belonging to the alkyl aryl polyether alcohol polymer group, formed from the polymerization of alkylphenol with formaldehyde and ethylene oxide.
Physical properties under normal conditions:
Viscous liquid, colorless to pale yellow, almost odorless
Specific gravity 1.03–1.06 g/mL at 25oC
Good water solubility
Soluble or highly dispersible in solvents such as ethanol, methanol, isopropanol, propylene glycol, glyceryl
Insoluble or very slightly soluble in mineral oil, paraffin, and saturated hydrocarbons
Slightly hygroscopic, requiring storage in airtight containers
HLB index: 13 – 14, acting as an O/W emulsifier
Tyloxapol is a commonly used excipient in pharmaceuticals, especially in respiratory preparations, ophthalmic preparations, and dispersion systems containing poorly soluble active ingredients. Besides, Tyloxapol can also be used as a mucolytic active ingredient in aerosol solutions due to its ability to reduce the viscosity of respiratory secretions. Currently, Tyloxapol is receiving more attention as a multifunctional excipient in the development of modern drug delivery systems, especially for eye drops, suspensions, emulsions, inhalers, and nano drug delivery systems.
2. Mechanism of action of Tyloxapol
Reduction of surface tension: similar to other surfactants, Tyloxapol has a structure consisting of a hydrophilic head and a hydrophobic tail, thus reducing the oil-water interfacial tension. This helps solid molecules like active ingredients to be wetted faster, increasing uniform dispersion and reducing the risk of agglomeration.
Micelle formation: the formation of micelles with a hydrophobic core containing hydrophobic active ingredients or poorly soluble solids inside, and a hydrophilic layer outside, helps increase the solubility and dispersion of solid molecules in the solvent.
Stabilization of dispersion systems: Tyloxapol adsorbs onto the surface of active ingredient particles, creating a hydration barrier and steric hindrance, which reduces collisions between particles, limits agglomeration, and maintains stable particle size throughout the storage period.
Reduction of respiratory secretion viscosity: in aerosol preparations, Tyloxapol reduces the surface tension between mucus and air, helping secretions to become thinner, reducing the adhesion of secretions to the respiratory mucosa, and supporting the drainage and expulsion of mucus. This is one of the properties that allow Tyloxapol to be used as an active ingredient in respiratory drug formulations.
3. Advantages of Tyloxapol surfactant
Non-ionic nature: Tyloxapol is a non-ionic surfactant, carrying no electrical charge, so it barely interacts with charged active ingredients or excipients, rarely forms complexes, and is minimally affected by environmental pH.
Stable operation over a wide pH range of 3 – 10
Low toxicity, high biocompatibility: suitable for use in eye drops and aerosol drugs.
Good dispersion and solubilization capabilities: helps dissolve and disperse poorly soluble active ingredients, slows down the agglomeration process, and stabilizes preparations.
Stabilization of nano systems: Tyloxapol is applied in nano drug delivery systems due to its ability to reduce particle size, prevent agglomeration, and maintain particle size distribution during storage.
Low foaming, no characteristic odor: convenient in formulation and usage.
Compatible with many active ingredients and excipients.
