This article details the polymer nature of ion exchange resins (IER) and their reversible complexation mechanism for effective taste masking. It highlights their safety, compatibility, and wide applications in suspensions, powders, chewable tablets, and ODTs.

Ion exchange resins (IER) are essentially insoluble high-molecular-weight polymers (typically a polystyrene matrix cross-linked with divinylbenzene) that contain attached charged functional groups:

When the ion exchange resin and the bitter active ingredient are mixed, an ion exchange reaction occurs at a specific pH. This is a reversible reaction:
R-COOH (ion exchange resin) + API (bitter) -> API-COOH (non-bitter complex)
The resulting complex is insoluble at the pH of saliva in the oral cavity; therefore, it does not interact with the bitter taste receptors on the tongue.
When this complex reaches the pH environment of the stomach or small intestine, decomplexation occurs, and the active ingredient is ready for release and absorption:
API-COOH -> API + R-COOH (ion exchange resin)
The ion exchange resin is subsequently excreted without affecting the pharmacokinetics of the active ingredient.
